Page 84 'FURTHER EXPLANATION' Section Example 1. Residential and nonresidential density calculations from the BDC Reference Guide (Updated August 2018), question:

1) Can someone please explain how to estimate the dwelling units (DU) within 400 meters? Is it estimated?

For the case of the example 1 within the reference, it is mentioned 680 DU within 400 meters.

In the example, the team apparently surveys the area to determine the number of dwelling units. Unfortunately, I don't believe GBCI will accept an estimate. I recommend searching for local tax records and/or maps for that information.
