We are working on a hotel project. The guest rooms have a key tag system where the lights and AC switches off as soon as the guest removes the key from the key slot. Can we adopt exceptional calculation methodology to claim savings under lighting and HVAC? Base case will be modelled with operating hours 24x7 and proposed case for about 16-18 hours working per day .

See Appendix D4 in the Advanced Energy Modeling Guide for LEED for a methodology that is acceptable for LEED. I do not think that your operating hour assumptions are reasonable.
