The project has breakout rooms with Interior glazing facing the exterior curtain wall, however, there is a column right in front of the interior side of the exterior curtain wall that affects the sightline. Does this count as an obstructed view? And does that mean the entire breakout room needs to be excluded from the area with views calculation?

Yes, columns are permanent and must be included in the sight line analysis. If the column blocks the entire view of the breakout room, then that space is 0% compliant.

The entire wall of the Breakout room facing the exterior curtain wall is all glass. The column does not block the entire view but a small portion.

So then only a portion of the breakout room is not compliant. Your sight line drawings should show you what area does and does not have access to views. Unless I'm misunderstanding what you are asking am of so private email me a drawing or image.
